The Protein Technologies Core Facility integrates five specialized Infrastructure Units that provide state-of-the-art technologies for the structural and functional characterization of proteins and their interactions.

Spanning proteomics, binding kinetics, structural biology, computational modeling, and multiplex protein profiling, the facility supports both experimental and in silico approaches.

Through expert support, advanced instrumentation, and integrated data analysis, it empowers high-impact research in life sciences, including biomarker discovery and therapeutic development.

INSTRUMENTS

The facility is equipped with:

  • Biacore X100 and T200
  • Tecan Spectrofluorometer
  • Biorad CFX Opus 96 PCR
  • SingleParticle Mainstream GPU Workstation
  • BIO-RAD Bio-Plex 200 System
  • Thermo Scientific Exploris 480 + Ultimate 3000 nano and micro
  • Ultimate 3000 biocompatible + DAD

Infrastucture units

The Core Facility is composed of five Infrastructure Units:

1. Proteomics

The unit is equipped with the state-of-the-art instrumentation for the analysis of proteins and peptides in biological samples.

SCIENTIFIC SUPERVISORS

The Unit offers:

  • Untargeted proteomic analysis with nano chromatography coupled to high resolution mass spectrometry of biological samples (biofluids, tissues, cells, etc.).
  • Qualitative and quantitative analysis of proteins and peptides.
  • Development of customized methods.
  • Support in experimental design.
  • Data analysis, graphical results, pathway analysis and bioinformatics.

2. Kinetic binding assay for the analysis of protein-protein and protein-ligand interactions

The unit is equipped with the state-of-the-art instrumentation for the analysis of kinetic parameters for the interaction between biological macromolecules.

The Unit offers:

  • Surface Plasmon Resonance analysis (Biacore X100 and T200).
  • Spectrofluorimetric assays (Tecan Spectrofluorometer).
  • Differential Scan Fluorimetry (Biorad CFX Opus 96 PCR).

3. Structure prediction of biomolecular interactions and docking analysis

The Unit provides computer-based structural prediction of protein-protein and protein-ligand interactions.

  • Structure prediction.
  • Affinity analysis.
  • Docking studies.

4. Structural Biology

The unit provide services for the high resolution structural investigation of biological macromolecules.

The Unit offers:

  • X-ray crystallography (access to ESRF syncrotrone): comprehensive support for protein crystallization and high-resolution structure determination. Unique in Piemonte region, the platform enables protein crystal production and diffraction data analysis, supporting advanced studies of protein–protein and protein–ligand interactions with excellent resolution and data quality.
  • Cryo-EM (SingleParticle Mainstream GPU Workstation): support for single-particle cryo-EM data processing and structural reconstruction. The facility hosts a high-performance GPU Workstation for large-scale data processing, representing the only in-house computational platform for cryo-EM data analysis currently available in Piemonte region.
  • Small-angle X-ray Scattering (access to ESRF syncrotrone): solution-based structural analysis of biological macromolecules, allowing the investigation of protein shape, conformational changes, and complex formation under near-physiological conditions.

5. Luminex technology platform for protein and cytokine profiling

The unit offers expertise in Luminex®️ technology for multiplex, simultaneous analysis of numerous analytes—such as cytokines, chemokines, growth factors, and signaling molecules—from small volumes of biological samples, including serum, plasma, and cell culture supernatants. This high-sensitivity method provides a broad dynamic range and supports biomarker studies, immune monitoring, and drug development
